Algul koosnes anatoomikum üksnes ringikujulise põhiplaaniga keskosast ehk rotundist, mis valmis Johann Wilhelm Krause plaanide järgi 1805. aastal. Hoone uhkeim osa oli amfiteatrina kujundatud valgusküllane auditoorium, kus tehti 1999. aastani õppetöö osana lahanguid.
Kuna anatoomikumi planeerides alahinnati arstiteaduskonnas õppida soovivate üliõpilaste arvu, tuli maja juba paarkümmend aastat hiljem laiendada. 1825.–1827. aastal ehitati rotundi külge kaks poolkaarekujulist tiibhoonet, mida pikendati 1856.–1860. aastal veelgi. Üliõpilased on aga sellest hoolimata meenutanud, kuidas õppeaasta esimesse loengusse tulles tuli neil oma initsiaalid ja kuupäev pingile kirjutada, et endale istekoht kindlustada.

The anatomical theatre originally only had the central part with a circular floor plan, or rotunda,
which was completed in 1805 according to the design by Johann Wilhelm Krause. The most
impressive part of the building was the light-filled auditorium, designed as an amphitheatre, where
autopsies were performed as part of the teaching until 1999.
As the number of students who wanted to study medicine was underestimated when planning the
anatomical theatre, the building needed to be enlarged already 20 years later. In 1825–1827, two
semi-circular wings were added to the rotunda and were further extended in 1856–1860.
Nevertheless, students remember that, when coming to the first lecture of the academic year, they
had to write their initials and the date on a bench to secure a seat.
